To provide simple methods for separating nucleic acid from nucleic acid-containing samples and simple methods for amplifying nucleic acid in samples.SOLUTION: The invention provides a method for isolating a nucleic acid comprising the steps of: contacting nucleic acid to phosphate and/or carbonate salt of alkaline earth metal of which solubility to water is 1 g/100 g H2O or less to form a complex; heating the formed complex of the nucleic acid and the phosphate and/or carbonate salt of alkaline earth metal salt at 45 degrees centigrade or higher.
