Provided is a method capable of purifying adeno-associated virus (AAV) contained in a sample under mild conditions using affinity chromatography. A purification method of AAV in a sample, comprising the steps of: Adding a sample containing AAV to an adsorbent containing an insoluble carrier and AAV-binding protein immobilized on the carrier to cause the adsorbent to adsorb the AAV; and eluting the AAV adsorbed on the adsorbent using a neutral buffer solution containing chloride ions or anions having a smaller Hofmeister series than the ions at 800 mmol/L or more.
