Provided is a method for inexpensively and conveniently constructing a highly diverse mutation library pool. The method for constructing a mutation library pool comprises the following steps (a) to (c): (a) a step for acquiring DNA fragments having 5' protruding ends, by using template DNA and a plurality of species of mutagenic primer pairs and carrying out a DNA synthesis reaction with each mutagenic primer pair using a DNA polymerase, wherein a mutagenic primer pair anneals with the template DNA, has a complementary region therebetween, and contains a mutation introduction site in at least one member thereof; (b) a step for causing a DNA ligase to act on the DNA fragments obtained in step (a); and (c) a step for transforming a host cell with the DNA obtained in step (b).
